The 52 -year-old actress, who plays PE teacher Sue Sylvester in the musical comedy-drama, announced the "difficult decision" to call time on her marriage in a statement to US publication, People on Monday (10 June).
"Lara and I have decided to end our marriage," she said.
"This has been a difficult decision for us as we care very deeply about one another. We ask for privacy as we deal with this family matter."
Jane and clinical psychologist Lara, 44, met at a fundraising event in San Francisco in 2009.
A year later, they tied the knot in an intimate ceremony held in Sunderland, Massachusetts - one of the few states in the US to allow gay marriage.
Jane, who is currently starring as Miss Hannigan in Annie: The Musical, also became stepmother to Lara's two daughters, Haden and Chase, from a previous relationship.
