Kasabian and Calvin Harris will headline the two-day festival, which is set to take place in August, while Sam and Ellie will also be performing for the crowds.
Former X Factor stars Olly Murs and Ella Henderson are among the lineup, along with The Script, Hozier, George Ezra, Stereophonics and Paloma Faith.
The festival, sponsored by Virgin Media, is pulling out all the stops to ensure fans are treated to the very best of British talent, as the event celebrates its 20th anniversary.
Simon Moran, festival director, says: "We wanted to champion British talent at this year's festival and, with the likes of Calvin and Kasabian leading the way, we don't think it'll disappoint.
"The eclectic mix of established artists and rising stars, with a few more in the pipeline, means that once again there is something for everyone at V Festival."
Speaking about the festival's history, he adds: "It's incredible to think what V has achieved in two decades.
"When we started out in '96, we were two stages and one tent, with the northern site only hosting one day of acts.
"Since then we've hosted some of the most iconic acts from around the world. Here's to another decade!"
V Festival takes place at both Weston Park, Staffordshire, and Hylands Park, Chelmsford, on 22 and 23 August. Tickets go on sale at 9am this Friday (6 March).
Music fans are advised to visit vfestival.com for a list of official, authorised ticket outlets.
