The TV favourites will front a new hour-long daytime series featuring celebrity interviews and features on "some of Britain's quirkiest real life characters."
Mel joked: "I've been asked to host a show with somebody called PORKIN? PARKIN..? PORTLY..? I've never heard of her - but I'll be very happy to meet her soon."
Hitting back, Sue said: "I very much look forward to working with Mel again, although, for legal reasons, we will now be at least one metre apart at all times."
The show has yet to be named but ITV has confirmed that 30 episodes will be made.
ITV Director of Daytime, Helen Warner, said: "I've been a big Mel & Sue fan for years... Both are brilliant broadcasters and comedians and bring a unique warmth and charisma as presenters. "
Mel and Sue can currently be seen hosting Great British Bake Off's new series, which airs Wednesday nights on BBC One.
